Karthik (Gopichand) belongs to Kadiyam, Rajahmundry, Andhra Pradesh. He works in a bank, and in the evening, Karthik becomes a coach of the village girl's Kabbadi team.
Karthik plays Kabbadi to fulfill his father's dream because his elder sister Bhoomi (Bhumika Chawla), whom he loves very much, wants him to play Kabbadi. The sole aim of Karthik is to represent Andhra State at the Women Kabaddi National Championship. He thus trains some talented girls from the village.
The girl's parents do not wish to send their girls to Delhi to play in the championship. However, Karthik convinces the parents and takes full responsibility for the girls. Soon Karthik and the girl's team start their journey towards Delhi to participate in the Women Kabaddi National Championship.
In Delhi, Karthik and the team meet Jwala Reddy (Tamannaah), coach for the Women Kabbadi team of Telangana, for the championship. Karthik and Jwala know each other Jawala has some misunderstandings, and thus, she leaves no opportunity to tease Karthik and his team. However, the Telangana team loses the match against the Punjab team and gets out of the championship. On the other hand, Karthik's Andhra team reaches in finals.
Karthik's sister Bhoomi (Bhumika Chawla) lives in Delhi with her loving husband, Aravind (Rahman), an honest, brave, and dedicated IPS officer. Bhoomi invites Karthik to celebrate the birthday of Aravind. Karthik wants to celebrate Aravind's birthday after the final match, but Bhoomi insists on coming with the Andhra Women Kabaddi team.
Karthik and the Kabaddi team reach there to celebrate their birthday. Aravind cooks himself to serve them. Girls start loud music and enjoy with Karthik, while Rahman cooks in the kitchen. Bhoomi also goes inside to help Rahman in the kitchen. At this time, some goon attack Aravind and start firing.
Aravind receives a bullet shot and is injured badly, while a good beats Bhoom, who plays smart by switching off the light so the music stops and Karthik learns about the attack. The trick works, and Karthik comes inside and compels the goons to flee.
However, Tiger Singh (Rohit Pathak) stays there to kill the entire family. Aravind throws a gun toward Karthik, and he shoots Toger to die.
The next day, while the girl's practicing in the stadium at 5 in the morning, some goons kidnap them. Karthik receives a call from Maakhan Singh (Tarun Arora), brother of Trilok. Maakhan asks Karthik to kill Aravind (Bhoomi's husband), or else he will kill the girls.
Jwala (Tamannaah) and her team help Karthik to know about Maakhan. Jwala introduces Karthik to a media friend, (Srikanth Iyengar), who helps to procure more about Maakhan Singh and his connection with Aravind. However, Maakan manages to kill the media person (Srikanth Iyengar) with the help of someone who belongs to the sports authority.
Maakhan gives only 48 hours to Karthik to kill Aravind (brother-in-law). And surprisingly, Laakhan is in jail and ruling from there.
What happens next? Will Karthiks kill Aravind to save the girls? Or does Karthik rescues his brother-in-law first? Who is Laakhan? What is the connection between Laakhan and Aravind? Will the girl play in the finals or not? The film Seetimaarr gives replies to all such queries.
