Manya Surve (Johan Abraham) is a loveable, innocent, and young final-year college student. He lives with his mother and elder brother, Bhargav Survey (Vineet Sharma).
Bhargav Surve (Vineet Sharma) is a petty criminal who indulges in illegal activities. Bhargav conflicts with an underworld don named Bhatkar (Ranjeet) and is currently on the run.
Vidya Joshi (Kangana Ranaut) is Manya's (John Abraham) love interest. Vidya insists upon introducing her to Manya's Mother. Manya finally introduces her to his mother on the day of his exam results.
While Manya brings Vidya home, Bhatkar's goons attack Bhargav. Manya does not wish to get involved in the fight, but Vidya compels him to save his brother. While Manya Surve grabs one of the goons, Bhargav stabs the goon, and he dies on the spot.
Manya flees from there and reaches college to check his results. However, a police Inspector, Ambolkar (Raju Kher), arrests him and publically humiliates Maya, who now lands in Yerwada Central Jail with his brother Bhargav (Vineet Sharma).
Potya (Chetan Hansraj), a prisoner, attacks Manya in jail. Munir (Tusshar Kapoor), another convict, saves Manya from the attack, but Bhargav succumbs to death during the assault.
Munir befriends Manya and suggests being strong and getting training from a comparatively more powerful prisoner, Veera (Sanjeev Chadda).
Veera trains Manya to become stronger. Potya (Chetan Hansraj) again attacks Manya from behind and beats Manya up, to which Manya responds by stabbing Potya to death.
Later, Manya and Munir make an escape plan and successfully flee from jail. They try to join hands with two brothers, Zubair Imtiaz Haskar (Manoj Bajpayee) and Dilawar Imtiaz Haskar (Sonu Sood), who rule Mumbai's deals.
However, a conflict arises between Manya and Dilawar, and Zubair disapproves of Manya joining his gang. After this, Manya forms a mafia gang with Munir and Veera. Soon Munir introduces Gyancho (Siddhanth Kapoor), a sharpshooter in the team.
First, Manya eliminates the local don Bhatkar (Ranjeet) and brings his bodyguard Jamal (Karan Patel) to his side. Next, Manya brings Inspector Ambolkar (Raju Kher) to his knees by beating him brutally in public.
Beating a police officer publicly enrages ACP Afaaque Baaghran (Anil Kapoor). He wants to catch Manya with the help of his team members, Inspector Raja Ambat (Ronit Roy) and Inspector Bhinde (Mahesh Manjrekar).
The story takes many twists and turns, and finally, Manya kills Zubair (Manoj Bajpayee). It enrages his younger brother Dilawar (Sonu Sood), who vows to kill Manya.
A gang war begins, and Dilawar starts killing the gang members of Manya. However, ACP Afaaque Baaghran (Anil Kapoor) has a unique plan to finish the gang war.
What happens next? Will Manya kill Dilawar, or Dilwar avenge his brother's death? What is the plan of Acp Afaaque Baaghran? What happens to Vidya? Film Shootout at Wadala gives replies to all such queries.
